Fully automated sample preparation and online HPLC analysis of CBD softgel capsules for potency testing
While more and more countries are in the process of legalizing cannabis, the demand for potency testing of cannabis products is growing. This application note describes a fully automated sample preparation and HPLC analysis method of Cannabinoids in CBD softgel capsules. A comparison of the results with a manual method demonstrates the improvement of recoveries and precisions while reducing hands-on times.
Summary
This application note describes the automation of sample preparation of 10mg CBD soft-gel capsules as well as the identification and quantification of cannabinoids such as CBD, THC and CBG. The method uses an accroma 2.0 sample preparation system interfaced with an Agilent 1260 Infinity II HPLC system to prepare and analyze the samples (samples IN – results OUT). The automated method was compared with manual sample preparation in terms of recovery, precision, and hands-on time. The recovery of automated sample preparation (127%) was higher compared to the manual method (102%). The precisions of the automated sample preparation were more than three times better for CBD, THC, and CBG compared with the manual sample preparation. With automation, the hands-on time per sample could be reduced by 54%. The results demonstrate the successful automation of the sample preparation of CBD soft-gel capsules and the added values of automation to laboratories in terms of accuracy, precision, and efficiency.
